Pengaruh Ekstrak Bawang Putih Tunggal (Allium sativum L.) terhadap Kadar Malondialdehid (MDA) Hepar Mencit (Mus musculus) Diet Tinggi Lemak

Abstract


Konsumsi makanan tinggi lemak secara terus menerus dapat meningkatkan akumulasi kolesterol dalam serum darah. Akumulasi kolesterol dapat meningkatkan stres oksidatif yang memicu kerusakan sel yang menghasilkan produksi reactive oxygen species (ROS). Di sisi lain, peningkatan jumlah ROS memicu peroksidasi lipid. Peroksidasi lipid menghasilkan produk dalam bentuk Malondialdehyde (MDA). Senyawa MDA bersifat reaktif dan mutagenik, berdifusi melintasi membran menyebabkan kerusakan biomolekul, yang mengarah ke berbagai penyakit metabolik kompleks lainnya. Kerusakan jaringan akibat radikal bebas dapat dicegah dengan antioksidan. Antioksidan dapat mengurangi produksi kolesterol dan mencegah radikal bebas. Bawang putih tunggal adalah rempah umbi yang memiliki banyak manfaat kesehatan dan mengandung antioksidan tinggi. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak bawang putih tunggal (Allium sativum L.) terhadap kadar MDA hepar pada mencit yang high fat diet (HFD). Dua puluh empat mencit Balb/C jantan dibagi menjadi enam kelompok: normal, HFD, HFD+simvastatin, HFD+Ekstrak bawang Putih Tunggal (EBT) 100 mg/kgBB, HFD+EBT 200 mg/kgBB, HFD+EBT 400 mg/kgBB. Pada hari ke 30mencit dibedahdan organ hati dikumpulkan. Kadar MDA diukur menggunakan metode Thiobarbituric Acid (TBA). Data kadar MDA dianalisis menggunakan Anova satu arah, kemudian dilanjutkan dengan tes DMRT. Hasil penelitian menunjukkan bahwa pemberian EBT memiliki efek signifikan pada kadar MDA hepar pada mencit yang diet tinggi lemak. EBT pada konsentrasi 100 mg/KgBB adalah dosis paling efektif untuk mengurangi kadar MDA hidup pada mencit yang HFD.
